歯周病原菌に対する防御性免疫応答の解析

研究概要	本研究目的は、歯周炎の病巣局所に増加する歯周病病原菌に対する体液性免疫応答の防御的役割の有無を検討することである。本研究では、まずそれらの歯周病原菌の表層抗原の精製を行い、それらに対する特異抗体を調整し、その特異抗体の防御的作用をin vitroの実験系および動物実験で検討した。歯周病原菌Porphyromonas gingivalis株の表層抗原である線毛、lipopolysaccharide(LPS)、莢膜多糖の抽出・精製を試みた。またActinobacillus actinomycetemcomitansの線毛を抽出精製した。Campylobacter rectusからはそのS-layer抗原及びLPSの抽出を行った。歯周病病原性細菌の上述抗原に対する特異抗体を調整した。ウサギ免疫抗血清およびS-layerに対する単クローン抗体は、オプソニン活性及び白血球の殺菌能を亢進させた。 A.actinomycetemcomitansに対する歯周病患者の防御抗体の検討から、本菌線毛抗原が感染予防ワクチン抗原になりうることを見い出すことができた。抽出した線毛抗原のアミノ酸配列を読みとり、その配列にもとづいたオリゴペプチドを合成した。合成オリゴペプチドは、MAP法によって分子量を大きくしてウサギに免疫し、線毛抗原に対する特異抗体を調整した。本ウサギ免疫血清は、in vitroの実験系で細胞やヒドロキシアパタイトビーズへの本菌の付着を抑制した。
