研究概要

1.尿キニン分解酵素(キニナーゼ)とその阻害薬
(1).ラット尿の主要キニナーゼはneutral endopetidase(NEP)とcarboxypeptidase Y-like exopeptidase(CPY)であることを確認し、ebelactone BがCPYを選択的に阻害することを確かめた。
(2).膀胱鏡を介して採取した患者の尿管尿においても、主要キニナーゼはNEPとCPYであることを確かめた。しかし尿pIIにより両者の活性比は左右され、酸性ではCPYの活性が強く、中性ではNEPの活性が強かった。従ってebelactone Bの作用は酸性側で強かった(原著8)。
(3).ラットのDOCA-食塩高血圧モデルでebelactone Bを持続皮下注入すると、尿中キニン量が増大し7〜10週までの昇圧作用が抑制された。
2.高血圧自然発症ラット(SHR)の腎カリクレイン分泌
(1).SHRでは離乳直後(7週令)より尿カリクレイン分泌がWistar Kyotoラット(WKY)に比し少なく、血圧のプラトーになる10週令で両者に差がなくなる。
(2).ペントバルビタール麻酔ラットにoxytocinを持続注入すると、WKYに比しSHRでは尿カリクレインの分泌量が著しく低い。しかしSHRの腎実質の腎カリクレイン含有量はWKYと変わらず、組織カリクレインの免疫組織化学的手法を用いても、oxytocin投与後に充分量が保留されていた。このことから、SHRは腎カリクレインの産生障害ではなく、分泌障害であることが示唆された。
(3).Naチャネルと結合している細胞骨格蛋白質のadducinの欠損の可能性を今後検討する必要がある。
