ムラサキツユクサを用いた環境変異原のin situモニタリングの基礎研究

研究概要

われわれの環境中に存在するさまざまな変異原については、環境中や試料中の濃度測定など、物理的あるいは化学的方法によるモニタリングが行われている場合があっても、実際の生物学的影響を検出する方法つまり現場での(in situ)生物学的とくに遺伝学的モニタリングは、その手法すら十分確立されていないのが現状である。
平成7年度の本研究では、花色について遺伝子型がヘテロ(青/ピンク)であるBNL4430株(アルキル化剤に高感受性)の若い花序つきshootsを培養液循環栽培装置内で大量に育成し、これを主な実験材料とした。
まず、各種変異原の効果の比較対象とするアルキル化剤については、前々年度のMMS、EMS、前年度のDMS、MNUに次いで、ENU(N-ethyl-N-nitrosourea)についても調査を行い、ENUが、X線との相乗効果を示さなかったMNUとは異なり、MMSと類似したX線との相乗効果を示すことをつきとめた。
前年度にX線と相乗的にも相殺的にも働くことを確かめた、除草剤として広範に用いられているプロミュータジェンのMH(maleic hydrazide)については、その植物細胞内での活性化がペルオキシダーゼによるものであることをつきとめ、X線と相乗的にも相殺的にも働く機構を明らかにした。
ガス暴露装置を用いて処理する大気汚染物質の変異原性の調査については、NO_2のO_2との反応を考慮し、N_2中でNO_2処理を行ったが、N_2中に置いた対照区でも突然変異頻度がある程度上昇し、目的とした低レベルでの実験は困難であった。空気中でのNO_2処理もNO_2の濃度によってNO_2/O_2比が変わり、一定した結果は得られなかった。現在、より容易と考えられるO_3処理をO_3発生装置を制作して実施中である。
