ポリアミンの生理作用と輸送機構に関する研究

研究概要	1.大腸菌の3種のポリアミン輸送系の1つであるプトレスシン排出系は分子量が46kDaの膜蛋白質(potE)で構成されており、その排出活性はプトレスシン・オルニチンアンチポーター活性に依存し、エネルギーを必要としなかった。このpotE蛋白質はプロトン駆動力をエネルギー源としてプトレスシンの取り込みも触媒することが明らかとなった。現在、排出と取り込みが同じ活性中心で触媒されているかどうか検討中である。また、この遺伝子発現がRNase IIIによるmRNAの5'-非翻訳領域の切断により翻訳レベルで活性化されることが明らかとなった。 2.動物細胞のポリアミン生合成律速酵素オルニチン脱炭酸酵素はアンチザイムという蛋白質と結合することにより、26Sプロテアソームにより分解される。このアンチザイムがポリアミン輸送系も負に調節していることを明らかにした。 3.動物細胞での蛋白質合成のポリアミンによる調節機序をウサギ網状赤血球の無細胞系で検討した。eIF-4EはmRNAのcap構造を認識し、mRNAの40Sサブユニットへの結合を促進する。このeIF-4Eによるcap構造認識はポリアミンにより促進をうけなかったが、eIF-4Eのリン酸化は促進をうけた。このことは、リン酸化によりeIF-4EのeIF-4Fへの変換が促進されるので、間接的にcap認識がポリアミンにより促進されることを示唆する。Met-tRNAのmRNA-40S複合体への結合はポリアミンにより強い促進をうけた。しかし、eIF4A,4Bによるヘリカーゼ活性はポリアミンで阻害された。mRNA中の5'-非翻訳領域のGC含量および長さはそれぞれ異なるので、このヘリカーゼ阻害の程度により、個々の蛋白質合成のポリアミンによる促進率が異なることが推論された。
