テトラサイクリン排出蛋白の分子構造と機能に関する研究

研究概要

テトラサイクリン排出蛋白推定膜貫通領域にただ一つ存在するHis257残基の役割について、部位特異変異導入法を用いて研究を行った。まず、His257をHis以外の19種類の全てのアミノ酸残基に置換した19種類の変異体を作成し、その性質を詳しく解析した。その結果、全ての置換体において、テトラサイクリン輸送活性は野性型の1/3以下に低下しており、257位残基がHisであることが湯粗鵜機能に重要であることが確認された。また、いくつかの変異体において、プロトンとの輸送共役が失われた脱共役変異体が得られたことから、この残基がプロトンとテトラサイクリンの輸送共役に関係する残基であることが示された。しかし、チロシンやメチオニンなどへの変異体がテトラサイクリン輸送能は低下するものの、共役の効率に有意の変化が無かったことから、イミダゾール環によるプロトンリレー機構の関与は否定された。また、トリプトファンおよびイソロイシン置換体のみ疎水性テトラサイクリン誘導体特異的輸送体に変化していたことから、この残基が基質の認識に関与することが示された。テトラサイクリン排出蛋白は分子内に全部で5個のHis残基を持っている。そこで次に、これらのHis残基を一つずつAla残基に置換した変異体5種類を作成した。その結果、テトラサイクリン輸送活性をほとんど失っていたのはHis257→Ala変異体のみであった。他の4変異体の輸送活性は野性型のものと変わらなかった。このことから、輸送機構に関与するHis残基は膜貫通領域のHis257のみであることが明らかになった。
