研究概要

ATP合成酵素(FoFl)は、呼吸鎖(電子伝達系)の形成するH^+の電気化学的ポテンシャル差を駆動力として、ADPとPi(リン酸)からATPを合成する酵素である。ATP合成酵素は可逆的な酵素でありH^+‐ATPaseとも呼ばれている。本研究はATP合成酵素による化学反応(ATP合成/分解)とH^+輸送の共役機構を分子レベルで明らかにすることを目的とした。
生化学的手法に加えて、本研究では変異の導入とその抑圧変異の分離という遺伝生化学的手法を駆使した。あるアミノ酸置換変異の効果が他のアミノ酸置換によって抑圧された場合、二つのアミノ酸残基が相互作用している可能性が高い。このアプローチを繰り返すことによって、近傍にあるアミノ酸残基を同定することができると考える。得られた主な成果は以下のように要約される。(1)本酵素の活性中心を持つβサブユニットの変異株および各変異を抑圧する変異株を系統的に分離し、活性中心を形成するGlu‐181、Arg‐182、Lys‐155、Thr‐156、Ser‐174などのアミノ酸残基を明らかにした。(2)H^+輸送路の一部を形成すると考えられるaサブユニットの膜を貫通する部分の変異株と対応する抑圧株を分離し膜貫通部分の構造とH^+輸送路を明らかにした。(3)ATP合成酵素を、通常の大腸菌膜のタンパクの30%まで大量生産する株を構築し、この株を用いH^+輸送路(Fo)を大量に精製し2次元結晶化の予備実験を終了した。(4)特にβサブユニットに注目し、γサブユニットとの相互作用を解析し、βArg‐52がγサブユニットのカルボキシル末端部分と相互作用をしていることを示した。(5)γサブユニット変異によりATP合成/分解とH^+輸送が脱共役している株を解析し、γサブユニットが共役の調節にプロトンゲートとして関与していることを明らかにした。
