血球分化に伴うビメンチン遺伝子の発現調節機構の解析

研究概要	マウス骨髄性白血病細胞M1は、IL-6やLIFといったサイトカインによってマクロファージへと分化を誘導される。この分化に際して、貪食能や運動能といったマクロファージに特有の機能が誘導されるだけでなく、細胞質の膨大や核の偏在と言った顕著な形態的変化をも誘導される。核がなぜある一定の細胞質領域に保持されているのかについては、まだはっきりとはわかっていないが、中間系フィラメントがその保持に関係していると考えられている。われわれはM1細胞の分化に際して、中間系フィラメントの一種、ビメンチンが顕著に誘導されてくることを見い出した。ビメンチン蛋白質の合成、その蓄積、ビメンチンmRNAの蓄積などをそれぞれ2次元電気泳動、ウエスタンブロット法、ノーザン解析などによって明らかにし、細胞内におけるビメンチン束の出現を、蛍光抗体法および電子顕微鏡による観察などによって明らかにした。またこの分化に伴うビメンチン遺伝子の誘導は、転写レベルで調節されていたことが、核ランオン解析によって明らかになった。そこでこの誘導調節機構を明らかにするため、ビメンチン遺伝子のプロモータ領域を解析することにした。マウス肝細胞より、ビメンチンのgenomic DNAをクローニングした。転写開始点より上流3kbまでの塩基配列決定を行い、ヒト、マウスおよびハムスターの間で、いくつかのエレメントがよく保存されていることを確かめた。CAT遺伝子をビメンチンプロモータの下流につなぎ、ビメンチンの発現に対して働くプロモータ/エンハンサーの同定を行った。その結果、M1細胞だけでなく、その他の繊維芽細胞においてもビメンチン発現を正および負に調節する2つのエレメントを同定することができた。
