研究概要	(1)セルトリ細胞による精原細胞の増殖促進作用:イモリ(Cynops pyrrhogaster)精原細胞ステージの精巣を解離し、密度勾配遠心によって精原細胞とセルトリ細胞を分離し、精原細胞のみ、あるいは、精原細胞とセルトリ細胞を混ぜたものを旋回培養し、〔^3H〕チミジンの取り込みによって増殖活性を測定した。精原細胞のみを±FSHで培養しても差はみられないが、セルトリ細胞を混ぜると+FSHの場合のみ増殖を活性化する。また、第一精母細胞ステージのセルトリ細胞を精原細胞に加えても+FSHで増殖を活性化しない。また、ミリセルを用いて精原細胞と同一ステージのセルトリ細胞をフィルターで隔てて培養してもFSHによる増殖促進効果はみられない。この結果は、セルトリ細胞の精原細胞に対する増殖促進作用はステージ特異的であり、互いの接着が必要であることを示唆する。(2)ペレット培養による減数分裂の開始:(1)の旋回培養系では精原細胞が1週間目に死んでしまったので、精原細胞ステージの精巣を解離し、再び遠心してペレットにし、コラーゲンマトリックスに埋め込んでフィルター上で培養した。-FSHでは大部分の精原細胞が死んでしまい、第一精母細胞へ分化しないが、+FSHでは2週間目で多くの細胞が第一精母細胞へ分化した。このような系は哺乳類では実現されていない。(3)イモリ精原細胞の増殖関連分子の検索:チロシンキナーゼの保存配列のプライマーを用いてイモリ精原細胞から抽出したRNAを鋳型としてRT-PCRを行ったところ、マウスのJAK1と88%の相同性を示すクローンを得た。in situ hybridizationを行ったところ、精原細胞の細胞質でシグナルが検出された。この結果は、イモリJAKが精原細胞の増殖機構において重要な役割を果たしていることを示唆する。今後このJAKを手がかりにして精原細胞の増殖機構を分子レベルで明らかにできる可能性がある。
