標的遺伝子置換法による脳機能の研究

研究概要	現代生物学の最も魅力的な課題の一つである脳機能の研究にとって、個体レベルの解析は不可欠である。最近、特定の遺伝子に突然変異を計画的に導入するジーンターゲッティング法(標的遺伝子組み換え法)が確立した。そこで、本研究班はこの方法を駆使して学習や記憶、知覚や認知などに関与する可能性のある遺伝子を破壊し、その形質転換を通して複雑多岐に亘る脳機能を分子に還元して研究しようとするものである。本研究はグルタミン酸受容体遺伝子に着目し、NMDAR1、R2A、R2C、およびmGluR2、mGluR6をそれぞれ計画的に破壊し、ホモ型突然変異体マウスの作成に成功した。mGluR6は、網膜に局在する。そこで、突然変異体に対して光刺激を与え、ON又はOFFに対するERGを測定すると、ONにのみまったく反応しないことが示された。このことはmGluR6が、ON刺激の伝達に直接関与しており、今後光刺激による学習についてこの分子の役割を研究できる材料が整った。また、ドーパミン受容体については古くから薬理学的実験が行われ、D1タイプとD2タイプが存在し、D2タイプは情動を介する記憶に関与していることが示唆されている。そこでD2受容体欠損マウスを作成した。その結果、D2受容体欠損マウスは、運動機能が不全で、パーキンソン病様の症状を示した。また、毛色が濃くなり、これは血中のα-MSHの濃度の20倍以上の上昇と関連していることがわかった。α-MSHは下垂体中葉から分泌されており突然変異マウスの中葉は過形成を呈していた。これらの事実はドーパミンD2受容体は、α-MSHの前駆体であるプロオピオメラノコルチンの発現を抑制しているだけでなく、細胞の増殖を抑制していることを示している。以上のことから、ジーンターゲッティングの脳研究への有効性が確認された。
