イノシトール三リン酸受容体ファミリーとシグナル伝達における生理機能の多様性

研究概要

IP_3受容体ファミリーを構成するタイプ1、2、3の分子構造、機能、発現分布の多様性について研究した。
1.ヒトからタイプ1、2、3受容体cDNAをクローニングし構造を決定した。
2.IP_3受容体の構造・機能相関について研究した。アミノ酸置換変異を導入した組換えIP_3結合部位を大腸菌で発現させ、[^3H]IP_3結合に関わるアミノ酸配列を解析した。カルモジュリンがLys1564-Arg1585の22アミノ酸と結合することを明らかにした。タイプ2も相同部位でカルモジュリンと結合するタイプ3は結合しなかった。アスパラギン結合型糖鎖結合部位は2475と2503の2つのAsnに存在することを明らかにした。部位特異的抗体を用いた免疫電顕で膜貫通トポロジーを検討し、6つの膜貫通断片とひとつのポア断片からなるチャネル構造モデルを提唱した。
3.各タイプmRNAのマウス末梢組織における発現分布をin situハイブリダイゼーション法で解析した。タイプ1は各種平滑筋細胞や第二次卵母細胞等に強く発現し、タイプ2は顎下腺小葉内導管の上皮、腎細尿管、輪卵管の上皮、卵巣の顆粒膜細胞と顆粒膜黄体細胞、精巣上体導管の上皮等に発現し、タイプ3は小腸上皮、唾液腺や膵臓の外分泌腺細胞、胃腺上皮等に発現していた。このことは、細胞種毎に多様なIP_3/カルシウムシグナリングを示唆している。今後、タイプ毎の細胞内の分布様式を解析する必要がある。
4.タイプ1遺伝子のプロモーターの制御でlacZ遺伝子が発現する組換え融合遺伝子(トランスジーン)を作製し、トランスジェニク・マウスを作製した。典型的なIP_3受容体の発現分布に類似したトランスジーン発現をする2系統を確立した。現在、発生分化や形態形成段階での発現パターンについて解析している。
