研究概要

1.ヒト単純ヘルペスウイルスのαトランス活性化誘導因子の結合ドメインとオーエスキー病ウイルス(ADV)前初期(IE)蛋白のDNA結合ドメインのキメラ蛋白がADVのIE遺伝子の転写を抑制すること、ならびにこれを発現する細胞がADV感染に対して抵抗することが判明した。この成績をin vivo のintracellular immunizationに応用するため、ヒトポリペプチド鎖延長因子EF-1αまたはマウスMx1遺伝子プロモーターで駆動されるように設計したキメラ蛋白遺伝子を調製した。これをマイクロインジェクション法でC57BL/6マウス受精卵に導入し、キメラ蛋白を発現するトランスジェニックマウスの作出を試みた。EF-1αプロモーターで駆動するキメラ蛋白遺伝子を導入した受精卵から計96匹のマウスが得られ、その中の1匹の体細胞に導入遺伝子を検出した。一方、Mx1遺伝子プロモーターで駆動するキメラ蛋白遺伝子を導入した受精卵から計33匹のマウスが得られ、その中の4匹に導入遺伝子が検出された。これらマウスから得たF1マウスの中で1系統のみに導入遺伝子が確認された。現在、このF1マウスの系統を確立することを試みている。
2.オーエスキー病ウイルス(ADV)の初期蛋白E90は、ADV遺伝子の転写を増強する。EP0の機能ドメインを決定するために、種々の欠失EP0遺伝子を作出し、これらが発現する蛋白の転写調節作用を検討した。その結果、EP0の転写増強作用にはRING finger領域を含むN末端から84番目までのアミノ酸配列および酸性アミノ酸領域を含む114から242番目までの配列が関与することが判明した。また、N末端から113番目までのアミノ酸残基からなるEP0変異体は、IEおよびTK遺伝子の転写を抑制し、EP0のdominant-negative mutantであることが判明した。
