新奇な海洋性超好熱細菌からの有用耐熱酵素の開発・利用に関する研究

研究概要	近年200℃以上に海底熱水口の発見とともに、100℃以上で増殖可能な新奇細菌群(超好熱細菌)が分離され、新しい遺伝子資源として脚光を浴びているが、高温・高圧といった培養条件の困難さから利用するには至っていない。本試験研究では、産業的に利用価値が高い80〜100℃で増殖可能な海洋性超好熱細菌を分離培養するシステムを確立するとともに、医学、遺伝子工学や食品工業の分野で切望されている超好熱酵素の検索、精製およびその遺伝子クローニングを用い利用しようとするものである。本年度の研究実績の概要は下記の通りである。 (1)既存の振盪回転培養機に耐熱性グリース、種々の断熱材や耐熱基盤を使うことにより、好気条件下で95℃まで振盪培養可能なシステムを確立した。本試作機を用いることにより、従来不可能であった好気性超好熱細菌を効率よく分離・培養することが可能となった。 (2)代表的な分離株としては、鹿児島県トカラ列島の子宝島の海岸熱水口より好気性超好熱細菌K1株、長崎県橘湾の海底熱水口より同細菌TB1株および同県小浜温泉より同細菌OH2株等である。いずれも好気性の不定型球菌で運動性を有し、至適増殖温度が90〜95℃であり従来の好気性細菌の至適増殖温度より10℃以上高かった。16SrRNA遺伝子の塩基配列より、これら3株は同属に属する近縁種で新種の原始的な古細菌であった。 (3)K1株は菌体外プロテアーゼを生産することから、本酵素を精製したところ分子量52kDaの金属プロテアーゼで、反応の至適温度は110℃でCa^<2+>存在下で著しく安定化され110℃4時間の加熱でも全く活性が低下しなかった。また昨年度分離した嫌気性超好熱細菌PyrococcusTS100株のDNAポリメラーゼは、その遺伝子クローニングを行い利用に向けて検討を行った結果PCR反応に有用であることが明らかになった。
