染色微粒子を用いた新しい血液型抗原の簡便で高感度な検査法の開発とその応用

研究概要

1)最近開発された青色及び白色アクリル系ポリマー微粒子に、血液型抗原及びその関連抗原と特異的に反応する抗体、レクチンを固定化し、それぞれの抗原の存在を着色された微粒子の凝集反応を通じて、高感度に特異的に検出する検査法が開発された。
2)種々のフコシル糖鎖抗原について、新規の反応特異性をもつ、マウスモノクローナル抗体が新たに得られたが、これらの中にはY,Le^b、Hタイプ2鎖とほぼ等しく特異的に反応するもの(YB-2)、Hタイプ1、2、3及び4鎖と等しく反応するもの(YB-3)など幾つかの有用な抗体が含まれている。これらを固定した着色微粒子によってそれぞれの抗原を高感度に検出することが可能となった。また、YB-2抗体は、大腸癌をはじめ幾つかの癌の組織化学的検索に有用であることを明らかにしたが、本研究の臨床応用の可能性が検討された。
3)抗体、レクチン類の代わりにBSA結合の種々の化学合成糖を固定し、その凝集反応によって抗体、レクチンの新しい検索が可能となった。
