新タイプの高血圧、動脈硬化治療薬としてのCNP関連薬剤の開発

研究概要	1.CNP遺伝子構造の決定: ヒト遺伝子ライブラリーより15kbのヒトCNP遺伝子断片をクローニングし、3kb BamHI-SalI断片の塩基配列を決定した。その結果、ヒトCNP遺伝子は少なくとも2つのエクソンと1つのイントロンより成ると考えられ、その5'隣接領域(1360 base pair)には、inverted CCAATボックス、GCボックス、cAMP応答因子様配列等を認めた。また、human-rodent hybrid DNA解析よりヒトCNP遺伝子が第2染色体に位置することが明らかとなった。更に、BALB/cマウスgenomic DNAより11kbのマウスCNP遺伝子断片を得、現在その構造決定を行っている。 2.培養血管内皮細胞からのCNP分泌調節因子の検討: 血管内皮細胞からのCNP分泌はtransforming growth factor-β(TGF-β)等の増殖因子に加え、種々のサイトカインにより調節を受け、特にtumor necrosis factor-α(TNF-α)により基礎値の100倍以上の分泌の亢進を認めた。またエンドトキシンそのものであるlipopolysaccharideも強力なCNP分泌刺激作用を示すことが明らかとなり、新しいペプチド性血管内皮由来弛緩因子としてのCNPの意義が示唆された。 3.血管平滑筋細胞/血管内皮細胞共存培養系の確立とCNP分泌機構の解明及び内皮由来CNPの意義の検討: 血管平滑筋細胞と血管内皮細胞の共存培養において、内皮細胞からのCNP分泌は著しく亢進し、分泌亢進した内皮由来CNPにより、血管平滑筋細胞でのcyclic GMP産生も上昇した。更にこの内因性CNPが血管平滑筋増殖に対し抑制的に作用することも明らかとなった。現在血管平滑筋細胞/内皮細胞連関におけるCNP分泌刺激因子を同定中である。 4.ヒト及びラット血中におけるCNPの検出: CNPに対し高親和性を有する抗CNPモノクローナル抗体の作製に成功した。これらの抗体を用いて、CNPに対する高感度ラジオイムノアッセイを確立し、ヒト及びラット血中にCNPが存在すること(各々1.4±0.6fmol/ml及び1.2±0.2fmol/ml)を初めて明らかにした。現在各種病態におけるCNPの分泌動態とその臨床的意義を検討中である。
