老人性神経細胞死を防止する興奮性アミノ酸関連薬物の設計と開発

研究概要	今までは代謝調節型グルタミン酸受容体の選択的アゴニストとして(1S,3R)‐ACPDというあまり強力でないアゴニストしか存在しなかったが、本研究により、L‐CCG‐I,DCG‐IV,cis‐およびtrans‐MCG‐Iなどの新しい代謝調節型グルタミン酸受容体アゴニストが見出された。ラット脳室内にDCG‐IV(100pmoles)を単回適用し、カイニン酸5mg/kgを全身投与すると、神経学的な症状観察では、カイニン酸による中枢興奮作用がある程度軽減するように思えた。カイニン酸の作用を確実に軽減させるために、生体内埋めこみ用浸透圧ポンプにより、DCG‐IVをラット側脳室内に持続注入(15時間)しておいて、そこにカイニン酸を2nmoles/rat側脳室に適用して、ラットの症状を観察したところ、痙攣発生を顕著に抑制するケースが認められた。更に、DCG‐IVの投与量を変動させて(24‐240pmoles/rat)、定量的に実験をおこなった結果、用量依存性にlimbic motor seizuresの発生を抑制することが判明した。しかも、極めて低い用量で有効であり、痙攣抑制にほぼ対応して、海馬CA3や扁桃核におけるカイニン酸誘発神経細胞死の発現頻度を有意に低減させた。バルビタールのように麻酔作用によって意識が喪失するような薬物が、カイニン酸による痙攣や神経細胞死を押さえることは既に判明しているが、意識の低下を伴わずにカイニン酸の激烈な中枢興奮作用を抑える薬物は少なくとも現在では極めてまれである。今までのグルタミン酸の生理作用に関する研究は主としてイオンチャネル型受容体を中心にして行われてきたが、このような作用を持つ代謝調節型グルタミン酸受容体アゴニストが出現したことは神経細胞死の今後の研究を進める上で画期的なことである。
