研究概要

DNAの塩基配列データやタンパク質のアミノ酸配列データから分子系統樹を推定するための最尤法の開発・整備をおこなった。塩基およびアミノ酸の置換確率行列を現実のデータから最尤推定し、これまでよりもはるかに現実的なモデルを構築することができた。最近ミトコンドリアDNAが分子系統学的な研究にひろく用いられるようになってきたが、ミトコンドリアでは遺伝暗号が核・細胞質系のものと多少異なるので、これまでミトコンドリアDNAの全塩基配列が明らかになっている脊椎動物20種のデータを用いて、ミトコンドリアDNAがコードしている蛋白質のアミノ酸置換確率行列を推定した。また、ミトコンドリアDNAのコドンの3番目の同義置換についても、塩基置換確率行列を推定した。このようなさまざまなモデルにもとづいて分子系統樹測定をおこなうプログラムを開発し、一般に公開した。
多重比較の観点からの最尤法によるトポロジー比較の研究を行なうとともに、分子系統学の著書において、方法論をまとめた。
このようにして開発したデータ解析法を、さまざまな生物学上の問題に適用して、いくつかの興味深い知見を得た。マグロ類8種の系統関係を調べた結果、ミトコンドリアDNAから推測される系統関係が、形態や核のDNAによるものと食い違った。このことから、ミトコンドリアDNAのイントログレッションの可能性が示唆され、特に近縁なもの同士の系統関係を推測する際には、注意が必要であることが明らかになった。
