C-C結合形成反応を触媒する耐熱性酵素による生理活性物質の新規合成法の開発

研究概要	好熱性細菌バチラス・ステアロサーモフィラスの遺伝子を大腸菌中で大量に発現させて得られるファルネシル二リン酸合成酵素を用いて、E、E-ファルネシル二リン酸を大量に合成する反応系を開発するための基礎を確立した。すなわち、Mg^<2+>イオンの存在下に高濃度の基質を仕込むと、反応生成物であるファルネシル二リン酸と無機ピロリン酸が共にマグネシウム錯体として沈澱するため、酵素反応が生成物によるフィードバック阻害を受けることなく進行するという反応工学的に極めて好都合な現象を見いだした。また、蛋白質工学的手法による研究の結果、この酵素のC末端領域にあるシステインとアルギニンはそれぞれセリンおよびバリンに置換しても触媒活性は保たれていること、またこれらの置換によりゲラニル二リン酸に対する親和性は変わらないが、イソペンテニル二リン酸に対する親和性が減少することなどが明らかになった。この親和性の減少は、ある条件下での生成物の鎖長の変化としても観察された。本酵素にある2つのシステインのうち1つはN末端に近い領域に、他方は領域C末端寄りの領域にあるが、前者は他の生物由来の酵素にも保存された場所にある。しかし、両システイン残基共に触媒機能に関与せず、いずれか一方、または双方のシステインをセリンに置換した3種類の変異体はいずれも野生型酵素と変わらぬ高い触媒活性を示した。これらのことは人為的変異の導入により、プレニルトランスフェラーゼの機能を変化させたり、新機能を付与したりすることが可能であることを示唆しており、有用物質合成への応用的新展開が期待される。
