中枢神経定量解析が画像データベース作成のための基本システムの開発

研究概要	本年度は共焦点レーザー顕微鏡による光学的切片を用いたstereolgyのdisector法を検討し、photo CDを利用した画像データーベースの試作に進めた。光学的切片を用いたdisector法は、核酸特異的蛍光色素propidium iodide染色切片を共焦点レーザー顕微鏡で、解析することで確立できた。開口数1.4のX60〜100の液浸系対物レンズを用い、Z軸方向に0.5-1.0μmステップで10-20枚画像を取り込み、取り込んだ画像を大型モニター上にタイル状に並べ、モニター上でdisector用のframeを各画像に載せることで効率良く解析できた。現在、スキャニングステージと組み合わせてdisector用の画像群を自動的に取り込むためのプログラムを検討中である。更に、中枢神経系で特定細胞グループの分布、密度が解析できる画像データーベースについても検討を進めた。50μm厚の切片を用い、蛍光免疫細胞化学によって特定の細胞グループを染色し、共焦点レーザー顕微鏡観察で、切片表面と切片中心部で陽性の細胞体の密度に差があるかどうかを検討し、両部位で差がない免疫細胞化学の条件を確立した。更に、蛍光免疫細胞化学後にABC法を適用し、蛍光抗体法で観察されていた全ての陽性細胞体が明視野で観察できる方法を確立した。免疫細胞化学で最も大きな問題となる抗体の厚い切片への不均一な浸透の問題が、このような厳密な検討で解決でき、目的であった特定の細胞グループの分布、密度が解析できる画像データベースのための標本が作成可能になった。現在、このような標本を写真撮影しスライドからphoto CDを作成したモニター上で定量解析を進め、顕微鏡下での標本からの直接解析と比較することで精度を検討している。
