核様体蛋白質H-NSによる遺伝子活性化機構の研究

研究概要	サルモネラの核様体蛋白質の一つであるH-NSは細胞内に約2万分子存在し、約400塩基対に1分子の割合でDNAに結合しており、転写や組換えなどの細胞調節機構において重要な役割を果たしているものと考えられている。本研究は、鞭毛オペロンであるflhDCオペロンの転写をモデル系として用いて、H-NSによる遺伝子活性化機構を分子レベルで解明することを目的として行われた。 1.flhDCオペロンのプロモーター領域の構造解明:flhDCオペロンの上流領域についてその全塩基配列を決定した。その結果、flhD遺伝子の上流には反対方向に転写される機能未知の142アミノ酸からなる蛋白質に対応する遺伝子が存在し、それとの遺伝子間には786塩基対のスペーサー領域が存在していた。この配列中にはH-NS結合部位に特徴的なAまたはTトラクトが豊富に存在しており、H-NSが優先的に結合するといわれている湾曲DNAが実際に存在することも確認された。 2.H-NS依存性の転写の同定:lacZとflhDCプロモーターの融合体を用いて、転写をβ-ガラクトシダーゼ活性で定量化する系を確立した。これから種々の欠失体を作成してその転写量を比較することにより、転写に必要なDNA領域を決定したところ、H-NS依存性の転写にはflhD遺伝子から500塩基対以上の上流領域が必須であることが判明した。一方、プライマー伸長法を用いてflhDCオペロンの転写開始点を決定したところ、この領域には4つの転写開始点が存在することが明らかになり、そのうちflhD遺伝子から530塩基対上流から開始される転写がH-NS依存性であることがわかった。
