免疫電顕法による核支配タンパク質分子のゴルジ体経由による葉縁体への輸送経路

研究概要	Brandtらは同調培養ユーグレナ細胞のLHCPIIタンパク質の合成はcell cycleの初期、中期に起こることを報告している。本年度の実施計画に示したように、同調培養ユーグレナ(Euglena gracilis Z)細胞を用いてはBrandtと同じように同調培養したユーグレナ細胞をもちいて、LHCPIIタンパク質分子の動態を追跡した。細胞はBrandtが行った同調培養法を用い、cell cycleにおける各ステージの細胞を経時的に採取し、LHCPIIの細胞内配置を急速凍結置換固定-連続切片法-免疫電子顕微鏡法により追跡した。LHCPIIはcell cycle中すべてのステージで、チラコイド膜上に認められた。一方、ゴルジ体に局在するLHCPIIはcell cycleの初期〜中期に限って観察された。すなわち、光照射直後の細胞を〓0時間細胞"とすると、LHCPIIのゴルジ体への局在性は全く見られない。〓3時間細胞"では60%のゴルジ構造にLHCPIIが局在し、〓8〜9時間細胞"ではおよそ95%に達した。その後、ゴルジ体への局在性は急激に減少し0%になる。次に、LHCPIIタンパク質分子の細胞内三次元分布を調べる為に、LHCPIIがゴルジ体に局在する9時間細胞の連続超薄切片像をもとに、コンピュータ・グラフィックスで解析した。9時間細胞のゴルジ体の個数は約6で、LHCPIIは細胞内の全てのゴルジ体に局在することがコンピュータ解析で判った。以上の結果から、新しく合成されたLHCPIIはゴルジ体を通過し、葉緑体に運ばれていることが新たに裏付けられた。
