免疫電顕法による核支酸タンパク質分子のゴルジ体経由による葉緑体への輸送経路

研究概要

先に免疫電子顕微鏡法により同調培養したEuglena細胞のcell cycle中および葉緑体形成過程における、光化学系II・集光性クロロフィルa/b結合タンパク質複合体(LHCPII)の局在を追跡した。その結果、LHCPIIタンパン質分子はゴルジ装置を経由し、葉緑体へ転送されることを最初に見いだした。今回、暗静置培養したEuglena細胞に0.1%ストレプトマイシンを添加し、正常な葉緑体形成を誘導する光を照度150 ft-cで照射し、葉緑体構造の形成過程を追跡するとプラスチド内部の構造はある程度の発達(チラコイド膜の形成等)が見られた。すなわち、プロラメラ体(PLB)に隣接し結晶様構造が形成されピレノイド構造から新しく5-6枚のチラコイド膜が形成された。しかしながら、ストレプトマイシン存在下では光照射後も、PLB構造の消失が起こらなかった。このような細胞について抗LHCPII抗体-免疫電顕観察すると、LHCPIIは結晶様構造上に局在しチラコイド膜には全く見られなかった。この結果から、ストレプトマイシン存在下でLHCPIIは細胞質で合成されゴルジ装置を経由しプラスチド内に輸送されるが、プラスチド内部のLHCPIIはチラコイド膜への転送が阻害されることが判った。すなわち、オルガネラ内部ではプラスチドで合成されたタンパク質がLHCPIIの輸送に関与していることが示唆される。
