液胞の物質輸送系の分子構築と生理的変動

研究概要	本研究は、植物細胞の中でも活発で多様な物質輸送がみられる液胞膜に着目し、その機能を支える輸送装置の分子構築とそれらの生理的な変化を解析することを目的とした。液胞は物質の集積のほか、無機イオン、代謝中間物の濃度調節、生体高分子の分解とリサイクル、細胞空間と充填するなど、多様な機能を担っている。これらの機能を支えているのは、液胞内を酸性に維持するプロトンポンプ、各物質に特異的なトランスポータおよびチャネルである。プロトンポンプは液胞膜に△_PH、膜電位を形成して他の二次能動輸送系に駆動力を与える役割も果たしており、液胞膜物質輸送系の最も重要な要素といえる。そこで、液胞膜の2種類のプロトンポンプ、H^+ATPaseとH^+ピロホスファターゼ(H^+PPase)を主な研究対象とした。H^+ATPaseは複数のサブユニットで構成される複合酵素であり、H^+PPaseは基質と酵素分子ともに単純であり、単一タンパク質で構成される。また、水チャンネルと考えられる疎水性タンパク質(VM23)についても検討対象とした。この研究により次の成果が得られた。(1)H^+PPaseは種子植物以外にも、シダ、コケなどに分布し緑色植物に普遍的な酵素であることが明らかになった。一方、水チャンネルVM23は、全ての植物に分布するわけではなく、シャジクモ、カサノリなどには存在しないことが明らかになった。(2)オオムギH^+PPaseの_CDNAをクローニングすることにより一次構造を解析し、単一タンパク質に基質分解の解媒部とH^+チャンネル形成部位の両方が存在することが解明できた。(3)種子が発芽する段階で、貯蔵器官のタンパク質顆粒は液胞化するが、この時、H^+ATPaseとH^+PPaseの両方が新たに合成され膜に局在化することが明らかになり、タンパク質顆粒-液胞の相互変換に明確な結論を与えた。このほか、細胞の成長、成熟にともなう2種プロトンポンプの量的変動についても解析し、両者が異なった変化を示すことが明らかになった。
