渦鞭毛藻の有毒株を識別可能なDNAプローブの開発に関する研究

研究概要	有毒渦鞭毛藻Alexandrium tamarenseおよびA.catenellaは麻痺性貝毒の最大の原因藻であるが、形態分類が困難なため発生のモニタリングに簡便で客観的な識別法が急務となっている。本研究は有毒鞭毛藻Alexandrium属の種間・種内ならびに有毒株をDNAレベルで識別可能なプローブを開発することを目的とした。その結果本年度は以下のような研究結果が得られた。 (1)日本各地の沿岸、タイおよび北米産のAlexandrium属6種24株の無菌・クローン株を用いて、昨年度の研究において制限酵素断片長多型解析により本属の識別が可能とされた5.8SrDNAと隣接するITS領域(内部スペーサー)をPCR法により遺伝子増幅を行い、クローニングした後に本領域の全塩基配列を決定した。これらの塩基配列(約520bp)を用いてJukes-Cantarの式により各株の遺伝子距離を求め、さらに近隣結合法によりその分子系統樹を作成した。その結果、A.tamarenseとA.catenellaは各々1つのブランチに入り両種を明確に区別することが可能であった。しかしタイ産A.tamarenseは、日本や北米産の形態学的同種とは遺伝的距離が大きかった。上記有毒2種は、無毒種であるA.affine、A.insuetumやA.pseudogonyaulaxとは明確に識別可能であることが明らかになった。 (2)ITS1の3'未満の149bpから177bpの塩基配列は、種内では同じであるが種間では多くの塩基置換が見られ、本属藻類の種特異的なマーカーとなり得る可能性が強く示唆された。そこで有毒種のA.tamarenseとA.catenellaに特異的な2種の蛍光でラベルされたDNAプローブを合成し、insituハイブリダイゼーションを行ったところ、両プローブで有毒2種の識別が可能となったことから有毒種に特異的なDNAプローブ開発の基礎が確立された。
