研究概要	プログラム細胞死はアポトーシスの過程を経て起こり、いわゆるネクローシスとは本質的に異なる細胞死の形態である。これら一連の現象にはストレス蛋白質が関与し、リンパ細胞の誘導や細胞蛋白質分解の促進などの生理作用が考えられている。その中でもアミノ酸76個で分子量8500の蛋白質であるユビキチンの存在が注目されている。本研究は、平成6年度を完成年度としているので、アポトーシス機構をDNAの変化と蛋白質の合成と分解を伴うものに絞って実験を遂行した。 1.グルココルチコイドがニワトリのリンパ組織の重量ならびに核DNAに及ぼす影響:合成グルココルチコイドの投与により、リンパ系の組織重量は減少し、免疫系臓器の退縮が確認された。さらに、これら臓器から抽出したDNAは、処理時間の経過と共に200bpを単位としてその整数倍の位置に梯子状に存在し、ヌクレオソーム間のリンカー部位でDNAの切断が確認された。これらの現象は、処理2時間後から認められ、時間の経過と共に鮮明になってきたが、12時間後からは次第に弱まり、リンパ細胞数の減少に伴うDNA量の減少を確認した。 2.リンパ細胞内における蛋白質量の変化:合成グルココルチコイド投与により、分子量19,000〜88、000の間で5種類の蛋白質の減少を確認した。他方、電気泳動像より分子量28,000,67,000ならびに80,000の位置のバンドが時間の経過と共に増加する傾向を示した。以上の結果より、減少した5種類の蛋白質はアポトーシスの抑制作用をもち、増加を示した蛋白質はアポトーシス誘導遺伝子の産物であることが示唆された。 3.ユビキチンの同定と動態:抗ユビキチン抗体を用いたイムノブロットを行い、ユビキチン量の変化を調べた結果、処理後6〜12時間までは増加を示したが、その後は減少傾向を示した。
