組織切片に含まれる細胞間マトリックスとそのmRNAの精密な定量法の開発

研究概要	実験的に作成した病態動物、ウイルス感染患者、健常な哺乳動物(家畜や実験動物)などから肝臓、腎臓、骨格筋などの組織を摘出し、液体窒素あるいはドライアイス・イソペンタン中にて急速凍結した後、クリオスタットを用いて未固定組織切片とし、この凍結組織切片に存在する細胞間マトリックス(ECM;I型コラーゲンなど)、速収縮ミオシン、それらのmRNAなどの生理科学的に重要な物質を検出・定量できる免疫組織化学的微量定量法を創出し、その有用性を確認した。従来からある生化学的定量法として比較して、(1)検出しようとしている物質の組織内での局在を保ちつ定量できる点、(2)原理的に免疫化学法を応用しているので1000倍のオーダーで高感度に検出できるので標本量が極微量ですむ点で優れている。病理学的場面では、組織細胞の形態をよく保存しながら様々な生理活性物質の精密な定量ができる本法を活用することで、正確な診断と予後予測が可能となる。また、高感度であるので、家畜や患者からバイオプシー標本のように極少量のサンプルを採取することで様々な生理活性物質の定量解析が可能であり、獣医畜産や臨床病理場面での様々な応用展開が期待できる。
