寿命を終えた腸上皮細胞の新しい処理機構

研究概要

1.モルモットの小腸絨毛先端部における上皮細胞の処理機構を解明した。寿命を終えた上皮細胞は、マクロファージと上皮内リンパ球の作用により管腔側細胞質を残して処理される。この“細胞の皮"は、両側の上皮細胞に押されて管腔側へもり上がり、細胞間隙が閉じた時に初めて管腔内に脱落する。この方式では、上皮バリヤーを破壊することなく寿命を終えた上皮細胞を排除することができる。
2.管腔内細胞成分の観察により、モルモットでは上皮細胞の本体は固有層のマクロファージに貪食されるため管腔内へ脱落しないこと、一方ラットやマウスでは核を含む胞体が管腔内に捨てられることが確認された。
3.モルモットの小腸絨毛の上皮内には、パーホリン陽性の果粒含有リンパ球が多数存在する。これらは上皮細胞と密接な関係にあり、細胞傷害性が強く、上皮細胞の細胞死(アポトーシス)を誘導することが示唆された。ラットにおいても上皮内リンパ球は豊富に存在するが、これらはパーホリン陰性であり、また上皮細胞を攻撃している兆候はなかった。ラットの場合、上皮細胞のアポトーシスは別の機構で調節されていると思われる。
潰瘍性大腸炎のモルモットのモデルでは、盲腸で粘膜のびらん形成が顕著であり、そこでは固有層のマクロファージは3倍程度増加していた。一方、ヒトの潰瘍性大腸炎は直腸に好発する。ヒトの正常腸粘膜では、貪食能を示すマクロファージは直腸粘膜に多いことからも、潰瘍性大腸炎とマクロファージの関係が注目される。
