慢性低酸素状態における動脈系化学受容器の分子機構と微細形態学的分析

研究概要

1.微細形態学的検討:長期間(12週)低酸素下(380torr)に暴露されたラット頸動脈小体(carotid body)の血管系は著しく拡張し、両生類の頸動脈小体(carotid labyrinth)と極めて類似した形態を示した。化学受容細胞と考えられているglomus cellを電子顕微鏡観察したところ、以下の様な新知見(微細形態学的特徴)が確認された。
(1)Glomus cellの周辺部は、支持細胞(supporting cell)に取り囲まれることなく、周囲の結合組織と直接接している。
(2)Glomus cellは細長い細胞質突起を血管周囲域へ伸ばしている。
(3)Glomus cellは血管内皮細胞や血管周皮細胞と直接接触している。
これらの微細形態学的特徴もまた両生類のglomus cellの特徴を示した。哺乳動物に比べ動脈血酸素分圧(PO_2)の低い両生類のglomus cellに類似した所見が得られたことは、これらの特徴が慢性低酸素状態に適応した結果であることを示すと共に、血液中からの酸素摂取ならびにカテコールミンの血中への放出を助長するに適した微細状態であると考えられる。
2.連続厚切り切片による三次元構築:微細形態学的検討に用いた試料より得た連続厚切り切片を超高圧電子顕微鏡(400KV)観察し、上記の特徴を立体的に把握することが出来た。さらに、glomus cellとglomus cell、glomus cellと支持細胞、およびglomus cellと内皮細胞の関係に着目し、正常環境下のものと比較検討した。特に、上記特徴(1)はglomus cellの肥大の割合が支持細胞のそれに比べ大きいことに由来して起こることが明らかとなった。本分析は、さらに例数を重ね検討を続けている。
これらの結果をもとに、神経支配を免疫組織化学的に検討し、さらに細胞内セカンドメッセンジャーを用いた生理学的検討をも行ない化学受容機構を総合的に解析しようとする次年度の研究結果が待たれる。
