グルタミン酸神経毒性に対する保護因子としてのコレシストキニンの作用機序

研究概要

コレシストキニンの神経保護作用機序を検討する目的で、ラット胎仔由来培養大脳皮質細胞におけるグルタミン酸神経毒性の機構、および、コレシストキニンの保護作用に連関する細胞内機構を検討し、以下の知見を得た。
1.NMDA受容体を介するグルタミン酸神経毒性は一酸化窒素ラジカルを介して発現することを明らかにした。さらに、カルシウム・イオノフォアのイオノマイシンが遅延性神経毒性を発現することを見出した。一酸化窒素合成酵素阻害薬、ヘモグロビン、スーパーオキシドディスムターゼ(SOD)などの一酸化窒素系を抑制する薬物は、NMDA毒性とイオノマイシン毒性をともに抑制した。一方、NMDA受容体遮断薬はNMDA毒性のみを抑制した。コレシストキニンはNMDA毒性とイオノマイシン毒性をともに抑制した。
2.コレシストキニンの神経保護作用はC-キナーゼ活性化作用を持つホルボール・エステルによって抑制され、A-キナーゼ活性化作用を持つフォルスコリンによって増強された。フォルスコリン、IBMX、8-bromo-cAMPなどの処置は、コレシストキニンと同様に一酸化窒素生成反応を抑制することにより神経保護作用を発現した。
3.Nerve growth factorおよびBrain-derived neurotrophic factorはNMDA受容体を介するグルタミン酸毒性に対して保護作用を発現したが、その効力はコレシストキニンと比較すると非常に弱いものであった。
これらの結果より、コレシストキニンの神経保護作用はNerve growth factorなどの神経成長、栄養効果とは異なる性質のものであり、その作用機序にはcAMP系の関与することが示唆された。
