胸腺の成長制御に果たす胸腺腫由来成長因子(MGFF)の役割

研究概要	(1)MGFFの精製:MGFFは99℃、5分処理によって失活するが、DNase,RNase,各種の多棟分解酵素やトリプシン処理に対しては安定であった。コンディションド・メディウムを濃縮し、DEAE陰イオン交換カラム、Sephacryl-S200ゲルろ過カラム、RCAレクチンカラム、C4逆相カラム、SDS-PAGEを用いて分画し、精製過程をSDS-PAGEを用いて解析したところ、トリプシン処理後の分子量約6万のタンパク質であることが推定され、ほぼ精製法を確率した。しかし、予想していたよりもMGFFの分泌量が低いために十分な量が集められずに本年度の目標であった蛋白の一次構造を明らかにしたり、抗体の作成までには至っていない。 (2)生物学的性質・精製されたMGFFにはほとんどCSF活性が見いだされなかった。また、脾臓細胞や骨髄細胞の中にもMGFFに反応する細胞が存在することが示された。精製された既知のサイトカインには活性や生化学的性質においてMGFFに相当するものは見い出せず、未知の因子である可能性が示された。また、新たにTaD-1-3細胞のコンディションド・メディウムに正常胸腺由来の上皮細胞株の増殖を阻害する因子が含まれていることが見い出された。 (3)ヌードラット胸腺上皮株の樹立:ラットヌード遺伝子の導入による胸腺腫発生の抑制の機序を探るために新たに無胸腺ヌードラット由来の胸腺上皮細胞株を樹立した。この細胞株は最近ヌードマウスやラットの原因遺伝子として報告された変異型whn遺伝子を発現しており今後この遺伝子の機能を解析していく上で非常に有用な細胞株になると思われる。今後、MGFF分取のための細胞株の再検討や分泌量をあげるための培養条件の設定を行い、速やかにMGFFの一次構造の決定を行いたい。一次構造を明らかにした上で、抗体の作成とcDNAクローニングを行い、本格的な胸腺の成長と胸腺腫の発生機序の検討を行う予定である。
