住血吸虫におけるホルモンリセプタースーパーファミリーの構造解析

研究概要

ファンクショナルクローニングからポジショナルクローニングへ、発想の転換と共に技法開発を取り入れて“寄生生物ゲノムプロジェクト"が始まった。住血吸虫を用いたYACライブラリーの作成と、FISH(Fluorescent in situ hybridization)による染色体マッピングは順調に進み、ゲノムサイズ2.7×10^8bpの生物に対し4.6×10^8bpカバー(約1.7倍)のライブラリーが得られ、それぞれの染色体特異的クローンのアセンブリにより、“contig"作成が可能となった。マーカーとして特に注目したステロイドホルモンリセプタースーパーファミリーは5'末端にY'Box binding protein領域をコードし、特殊な転写調節を有する。1.2kbフラグメントの遺伝子配列が決定され、YACライブラリーのスクリーニングによりマクロマップ・染色体マッピングが進行中である。なお、先年までのcDNAライブラリーランダムシークエンスによるESTのジェネレーションにより、Venter主催のEST Gene/Bankへの当録も既に完了した。寄生虫種の“疾病関連遺伝子群"のスクリーニングが開始された。
