研究概要

本研究では、1)下等脊椎動物からMHC(主要組織適合遺伝子複合体)遺伝子をクローニングすることにより、MHC遺伝子の系統発生学的起源を明らかにすること、2)軟骨魚類から哺乳類におよぶ多様な生物種のMHC遺伝子を解析することにより、MHC遺伝子の長期進化を規定する一般的な法則を明らかにすること、を目的とした。まず第一番目の目的に関しては、軟骨魚類には、多型性に富んだ典型的なMHC遺伝子が存在することを明らかにした。しかし、それより下等な脊椎動物である無顎類からMHCクラスI,II遺伝子を分離しようとする試みは、現在のところすべて失敗に終わっている。したがって、無顎類にはMHC遺伝子は存在しないか、あるいは、存在するとしても他の脊椎動物のMHC遺伝子とはかなり配列が異なっているものと推定された。最近、我々は、軟骨魚類にはプロテアソームのサブユニットをコードするLMP7,LMPX両遺伝子が存在するのに対し、無顎類にはLMPX遺伝子しか存在しないことを示唆するデータを得た。これらの結果を総合すると、無顎類には典型的なMHC遺伝子が存在しない可能性が示唆された。また、MHC遺伝子の長期進化を規定する一般的な特徴として、以下の3点を同定した。1)MHC遺伝子では、種間配列保存の程度が低いにも関わらず、全体的なドメイン構造はよく保存されている。2)機能的に重要なMHC遺伝子の数はヒトからサメにいたるまで大差なく、ハプロイドあたり1〜3個の範囲にある。3)MHC遺伝子多型の本質的特徴は種をこえて温存されている。
