アデノウイルスベクターを用いたIL-6遺伝子導入による間質性肺炎モデル動物の作製

研究概要	Wisterラットに10^8pfuのlacZ遺伝子組み込みアデノウイルスベクターを経気道的に導入した結果、生理的条件下では増殖性に乏しいII型肺胞上皮細胞や肺胞マクロファージで、少なくとも3週間にわたりlacZ遺伝子発現が持続した。つぎに1.8×10^8pfuのSRαプロモータ支配ヒトインターロイキン6(IL-6)、ヒトIL-6受容体(IL-6R)遺伝子組み込みアデノウイルスベクター、あるいはその混合を経気道的に投与し、それぞれの遺伝子発現とラット肺の病理組織学的変化を検討した。アデノウイルスベクター投与72時間後にはIL-6とIL-6Rの混合投与によって著明なリンパ球浸潤を伴う胞隔炎を認め、さらに肺胞腔内に肺胞マクロファージ(AMφ)の浸出を認めた。一方IL-6あるいはIL-6Rの単独投与やサイトカイン遺伝子を持たない対照ベクター投与ではなんらの組織学的変化も認めなかった。IL-6とIL-6R遺伝子組み込みベクターの混合投与によって惹起された胞隔炎の部分に一致して、IL-6およびIL-6R遺伝子発現がin situ hybridizationで、さらに同タンパクの発現が間接蛍光抗体法や免疫組織染色法でそれぞれ確認された。しかし、病変部分のAMφと一部のII型肺胞上皮細胞のみがこれらのタンパクを強発現しており、肺胞壁に浸潤したリンパ球にはほとんど発現を認めなかった。したがって導入されたIL-6とIL-6Rがラットの信号伝達タンパクを使って宿主細胞に何らかの変化を惹起し、これらの病理学的変化を引き起こした可能性が高いと考え、この病態の解明を試みている。さらにサイトカイン遺伝子導入後長期および頻回投与後の病理学的変化についてもあわせて検討を加えている。
