アルツハイマー型老年期痴呆における流血中神経受容体抑制因子の同定と病因的意義

研究概要	ムスカリン性アセチルコリン受容体に対するアルツハイマー型老年期痴呆例流血中の阻害因子の詳細につき検討を進めた。血清中に存在するリン脂質の一種であるlysophosphatidyl cholin(LPC)はin vitroにおいて濃度依存的にラット脳シナプス膜分画ムスカリン性アセチルコリン受容体に対する[^3H]quinuclidinyl benzilate.([^3]QNB)結合を抑制する。しかし他のリゾリン脂質であるlysophosphatidylethanolamineやlysophosphatidyl serineにはこの抑制作用は認められなかった。またLPC(各10^<‐4>M)の1位の脂肪酸側鎖がC10:0のdecanoyl基位下の短鎖脂肪酸より成るLPCでは[^3H]QNB結合抑制の程度は10%以下と低値であったが、C14〜C18の飽和脂肪酸であるmyristoyl基〜stearoyl基より長鎖の脂肪酸よりなるLPCでは[^3H]QNB結合抑制は30%前後と高値を示し、さらに不飽和脂肪酸であるoleoyl(C18:1)、linoleoyl(C18:2)、arachidonyl(C20:4)ではさらに高度の抑制を示した。またLPC10^<‐4>Mの[^3H]QNB結合抑制作用は10^<‐3>Mアルブミン添加によるほぼ完全に抑制され、血清中では大部分のLPCがアルブミンと結合していると考えられている。しかし、アルツハイマー型老年期痴呆群では血清アルブミン値は有意に低下しており、アルツハイマー型老年期痴呆例では血清中遊離型LPC濃度の増加が、脳シナプス膜ムスカリン性アセチルコリン受容体に対する阻害作用を示している可能性がある。以上、アルツハイマー型痴呆例の流血中ムスカリン性アセチルコリン受容体阻害性因子の一因子として流血中LPCがあげられ、老年期痴呆に関与している可能性を示した。
