研究概要

エポレセプターからの細胞増殖刺激伝達に重要な領域を同定する目的でレセプターの細胞内領域で他のサイトカインレセプターと相同性を有する膜近位部に数種類のアミノ酸置換を導入した変異エポレセプターを培養細胞に発現させ検討を行い、この部位の保存されたアミノ酸配列がレセプターのチロシンリン酸化誘発能、c-mycやc-fos等の遺伝子の発現及び細胞増殖刺激伝達能に必須であることを明かにした(Mol.Cell.Biol.13:1788-95,1993)。
エポレセプターからのチロシンリン酸化を介した細胞内刺激伝達を司るチロシンキナーゼを同定する目的で造血系細胞に発現される種々のチロシンキナーゼにつき検討を行い、その内の一つのJAK2キナーゼがエポ刺激によりエポレセプターの上述の細胞内膜近位部と結合し活性化される事を見いだした(Cell74;227-236,1993及び現在投稿中)。
チロシンリン酸化されたエポレセプターと結合し刺激情報を伝達する因子の存在を検討する目的でSH2(src homology2)領域を有する幾つかの因子につき分析を行いphosphatidylinositol 3-kinase(PI3K)がエポレセプターと結合することを見いだした。種々の変異レセプターを発現させた細胞での検討よりこの結合にはチロシンリン酸化されるレセプターのC末端領域が必要であること及びこの結合はエポレセプターからの増殖刺激伝達に必須ではないことを明かにし得た(J.Biol.Chem.269:614-620,1994)。
