エリスロポエチン遺伝子の発現調節機構-合成オリゴヌクレオチドのgene-targeted drugとしての開発と遺伝子操作

研究概要	エリスロポエチン(Epo)遺伝子の調節配列の解析は、その活性が低く不明であった。我々は三重鎖形成を応用したantigene strategyを用いた解析を行った。つまり合成オリゴヌクレオチドを、Epoを産生するHep3B細胞に添加してEpo遺伝子の調節配列で三重鎖を形成させると転写因子の調節配列への結合が阻害され転写レベルに変化が起きる。この変化をcompetitive polymerase chain reactionによりEpo mRNAを定量して解析した。この結果、Epo遺伝子5′上流の転写開始部位より-60bpのCACCC配列に対するantigene投与でEpo mRNA産生量は用量依存性に低下し、CACCC配列は生の調節配列であることを確認した。-30bp上流のGATA配列に対するantigeneを投与すると用量依存性にEpo mRNA産生が亢進し、GATA配列は負の調節配列であることを確認した。また電気泳動で上記領域の三重鎖形成を確認した。さらにゲルシフトでCACCC-GATA配列に特異的に結合する転写因子を認めた。CACCCの転写因子はゲルシフトにおけるbandのintensityが低酸素で増加するため誘導因子と考えられる。一方GATAの転写因子は、そのintensityが低酸素で減少するため抑制因子と考えられ、以上を報告した(J.Biol.Chem.1994)。次にこのGATA配列に結合する転写因子の同定を行った。まずHep3B,HepG_2細胞の発現するmRNAをNothern blottingでGATA-1,2,3因子をプローブとして解析すると、ともにGATA-2 mRNAを発現していた。またモノクローナル抗体を用いた細胞化学染色でも、ともにGATA-2蛋白の発現を確認した。さらにHep3B細胞へ外因性にGATA-2 DNAを遺伝子導入すると、Epo mRNA産生が低下することを認めた。以上より、Hep3B細胞内ではEpo遺伝子のプロモーター、GATA配列にはGATA-2因子が結合し負に発現調節していることが認められ、報告予定である。
