金属ポルフィリンおよび遺伝子操作を利用した新生児黄疸治療法の開発的研究

研究概要	Tin-protoporphyrin(Snpp)による光誘導性生体障害機構の解明を続けた。長江はSnPP-光照射ラットについて血液成分の経時的な変化を検索し、急激な血糖値の低下が起こる(正常範囲の1/10以下に達する)ことを見いだした。これはSnPP-光照射ラットが高率に死亡する原因の一つであろう。慶野と坂野はSnPPの光増感性の軽減を目的としてスカベージ効果を持つ薬物を検索し、L-ascorbic acidがin vitro,in vivoともに有効であることを見いだした。また、SnPPの持つ最大の欠点である光増感性を持たない金属ポルフィリンの合成を計画した。中心金属のイオン化傾向がSnより低く、ポリフィリンの側鎖が自由電子を受け取りやすいことを念頭に合成した金属ポリフィリンの内で、cobalt-mesoporphyrinはhemeoxygenase活性阻害効果を持ち光増感性は全く示さないかった。青野を中心にCrigler-Najjar症患者の持つbilirubin:UDP-glucuronosyltransferase(BRUGT)遺伝子の解析を進めた。I型患者はBRUGT遺伝子の第1エクソン部分に1塩基欠失が起こりストップコドンが現れ、BRUGTタンパクが作られないことが原因であった。II型患者はBRUGT遺伝子の塩基置換が起こっていた。置換の位置、種類は患者によってそれぞれ異なっていた。塩基置換が起こることでその位置のアミノ酸が変わり、BRUGT酵素タンパクの立体構造が変わりビリルビンにグルクロン酸を抱合する機能が低下したものと推測された。患者はそれぞれの遺伝子変異に関してホモであった。
