胎児外科的に作製された無嗅脳症マウスの脳形態形成異常

研究概要	無嗅脳症では、しばしば嗅球欠損と脳梁欠損が合併することが報告されてきた。また、我々が系統維持している無嗅脳症/多脂症マウス(Pdn/Pdn)でも嗅球欠損のほか、脳梁欠損、前交連、水頭症などを合併する。そこで、嗅球が欠損することと脳梁欠損を代表とする脳の異常が独立して発症するものではなく、何らかのシークエンスを持っているのではないかと考え、そのシークエンスを解きほぐる事を本実験の目的とした。マウスの子宮切開法を用いて、卵巣嚢を通してレーザー光を胎仔の嗅球原基に照射すると、その後の脳形態形成において、左右の脳梁線維が交叉せず、また、視床や前交連の異常も認められた。さらに、胎生期を遡って検討した結果、脳梁線維が交叉する場である交連塊に異常を認めた。遺伝性無嗅脳症マウス(Pdn/Pdn)の胎生15・16日でも、交連塊が欠損していた。一方、正常マウスでは、胎生12日までに嗅神経線維が終脳に接着し、13日には、嗅球が終脳から誘導される。ところが、Pdn/Pdnマウスの胎生12日の脳を観察すると、嗅神経線維が終脳に接着していなかった。以上の事から、嗅神経線維が終脳先端に接着しない事が嗅球欠損を惹起し、この嗅球欠損が脳梁欠損や前交連の異常を惹起するという発症シークエンスが解明された。 Pdn/Pdnマウスでは、水頭症を合併し、多くのアポトーシスが観察される。そこで、このアポトーシスが原因で水頭症が合併しいるものと想定し、アポトーシスに関連すると云われているTRPM-2遺伝子をプローブにノーザンブロットを行った結果、Pdn/Pdnの脳幹部で強い発現が認められた。
