研究概要	無嗅脳症では、しばしば嗅球欠損に伴って脳梁欠損や水頭症が合併することが報告されてきた。また、我々が系統維持している遺伝性無嗅脳症/多指症マウス(Pdn/Pdn)でも嗅球欠損のほか、脳梁欠損、前交連欠損、水頭症を表わす。そこで、嗅球が欠損することと他の脳の異常が独立して起きるものではなく、なんらかのシークエンスを持っているのではないかと考えてきた。嗅球に層構造を構築する僧帽細胞は、嗅神経の支配を受ける。Pdn/Pdnマウスでは、嗅神経線維が中枢神経系に接着せず、嗅球が欠損している。従って、僧帽細胞が欠損し、さらに嗅皮質である梨状葉皮質も欠損している。そこで、本年度は、嗅球欠損に伴って、僧帽細胞がどこへ行ったかを調べた結果、胎生14日には僧帽細胞と、その神経支配を受ける梨状葉皮質の細胞がアポトーシスを起こしていることが判明した。このことを、さらに実験的に調べるために、胎生13日の正常マウス胎仔の嗅球原基をレーザー光で破壊し、無嗅脳症にすると、胎生14日の梨状葉皮質にアポトーシス細胞が認められた。さらに、片側の嗅球だけを破壊すると、胎生18日の胎仔では、嗅球の残っている側の梨状葉皮質は形成されていたが、嗅球が欠損した側の梨状葉皮質は欠損していた。以上のことと、昨年度までの研究結果から、嗅神経線維が終脳に接着しないことが嗅球欠損を惹起し、この嗅球欠損が脳梁欠損や前交連の異常を惹起する。さらに、嗅神神経の支配を失った嗅球の僧帽細胞はアポトーシスを起こし、嗅球の僧帽細胞の神経支配を失うと梨状葉皮質は形成されないという発症シークエンスが証明された。
