口腔発癌モデルを用いた口腔癌発生の分子病理学的追求とその化学予防の基礎的研究

研究概要

これまで4-Nitroquinoline 1-oxide(4-NQO)誘発ラット口腔発癌モデルを用いて、このモデルにおける口腔癌発生の分子病理学的背景の検討と柑橘類等に存在し抗酸化作用を有するプロトカテク酸(PCA)を含むいくつかの化合物の口腔発癌抑制作用を報告してきた。本年度は、口腔発癌プログレッションモデルの確立とPCAのプログレッション抑制作用の有無を検討した。
<材料と方法> 雄F344ラット(6週齢)88匹を5群に分け以下の処置を行った。第1群:4-NQO(20ppm)を4週間飲水投与。第2群:第1群の処置に加えて、実験開始後18週より2週間のサイクルで4-NQO(20ppm)を計4回飲水投与。第3群:第2群の処置に加えて実験開始後16週より16週間0.2%PCAを混餌投与。第4群:0.2%PCAのみを16週間混餌投与。第5群:無処置対照群。実験は32週で終了し、剖検後口腔を含む諸臓器の病理学的検索を行った。
<結果> 口腔(舌)腫瘍は第1-3群に発生し、乳頭腫・扁平上皮癌の発生頻度は第1-3群でそれぞれ16%・0%、33%・47%、10%・10%(1匹に肺転移)で、第2群、第3群の扁平上皮癌の頻度はそれぞれ第1群、第2群に比し有意に増加、低下していた。
<結論> 以上の結果より、本モデルは口腔癌のプログレッションモデルとして有用であること、またPCAはそのプログレッションを抑制することが示唆された。
