高転移性ヒト唾液腺癌細胞株の樹立とその転移機構の解析に関する研究

研究概要	前年度の研究実績より,非転移性ヒト唾液腺癌細胞株が転移性唾液腺癌細胞に悪性転換するin vitroでのシステムは樹立されたものと考える。従って当該年度においては、上記のシステムを用いてその転移機構に包含されているメカニズムの解析を行なった。一般に癌の局所での増殖から浸潤・転移までのすべての過程において不可欠であるとされる腫瘍関連血管新生については、癌細胞から分泌される血管新生因子が重要であると考えられている。そこで、癌細胞から分泌される血管新生因子とその役割につき解析し、以下に述べる結果を得た。1.転移細胞クローンの培養上清(CM)中にはウシ大動脈由来血管内皮(BAE)細胞の増殖と遊走を促進する因子の存在することが判明した。2.CM中にはBAE細胞の蛋白分解酵素産生を抑制する因子の存在が確認された。そしてこれら1.と2.の作用を有する因子が上皮成長因子(EGF)であることを明らかにした。3.非転移性唾液腺癌細胞クローンに比較して転移細胞クローンはEGFを多量に分泌するにもかかわらず、EGFレセプター数に関しては同程度であった。4.さらに、EGFは癌細胞自らの転移能には影響を及ぼさず、BAE細胞に選択的に作用することが明らかとなった。従って、以上の所見より、転移性ヒト唾液腺癌細胞は非転移性癌細胞に比較して血管新生因子であるEGFを多量に産出・分泌し、しかも分泌されたEGFは自らの転移能には影響を及ぼさず、血管内皮細胞に選択的に作用して血管新生を促進することが明らかとなった。すなわち、EGFの作用はパラクリン機構を介して作用することが判明した。
