ポリアミン輸送系の分子生物学的解析

研究概要

1.大腸菌のポリアミン輸送系は3種存在するが、そのうち1つはスペルミジンの優先取り込み系であり、基質結合蛋白質(potD)、膜貫通蛋白質(potBおよびC)、およびATP結合部位を有する膜表在性蛋白質(potA)の4種より構成されている。このスペルミジン輸送欠損株の変異部位を解析したところ、potA蛋白質のVal-135がMetに置換されていた。この変異potA蛋白質はATPとは結合できたが、ATPaseが消失しており、この部位がATPaseの活性中心であることが推論された。また、8-アジドATPを用い、ATP結合部位を探索したところ、8-アジドATPはCys-26に結合していた。以上の結果より、ATPaseの活性中心はN末端側に存在することが明らかとなった。
2.ポリアミン輸送系3種のうちの第2は、プトレスシンの排出に関与するpotE蛋白質である。この蛋白質は分子量46kDaで、12ケ所の膜貫通部位を有する膜貫通蛋白質である。プトレスシンの排出はオルニチン・プトレスシンアンチポーター活性に基づいており、エネルギーを必要としなかった。potE蛋白質は排出活性に加え、プロトン駆動力をエネルギー源としてプトレスシンの取り込み活性も有することが明らかとなった。現在、排出と取り込みが同じ活性中心で触媒されているかどうかsite-directed mutagenesisの手法を用い解析中である。また、この遺伝子の発現は酸性条件下活性化されるが、この誘導にはRNase IIIによるmRNAの5'-非翻訳領域の切断が関与しており、少なくとも活性化の一部は翻訳レベルで起こることが明らかとなった。目下、転写レベルで働く活性化因子の遺伝子をクローニング中である。
