ポリアミン輸送系の分子生物学的解析

研究概要

1.大腸菌のスペルミジン優先取り込み系は基質結合蛋白質(potD),膜貫通蛋白質(potBおよびC)、およびATP結合部位を有する膜表在性蛋白質(potA)の4種より構成されている。このpotA蛋白質の機能をX線結晶解析およびsite-directed mutagenesisにより変異蛋白質を作製し、解析した。その結果、N末端側にATP結合部位とATPase活性中心が存在し、C末端側は膜貫通蛋白質との相互作用に関与していることが明らかとなった。同様な手法を用いて、potD蛋白質のスペルミジン結合部位の同定を試みた。その結果、Glu36とGlu171が一級アミンとの相互作用に、Asp257が二級アミンとの相互作用に関与していることが明らかとなった。
2.プトレスシンの排出に関与するpotE蛋白質(プトレスシン・オルニチンアンチポーター)の活性中心を、この蛋白質に存在する16個の酸性アミノ酸(GluまたはAsp)を中性アミノ酸(GlnまたはAsn、およびAla)に変換することにより解析した。その結果、Glu207が活性中心であることが明らかとなった。そのほかにGlu77、Glu113、Glu409が活性に関与していることが示唆された。
3.細胞内ポリアミン量は生合成と輸送の両者に依存している。動物細胞の場合は生合成の律速酵素であるオルニチン脱炭酸酵素(ODC)の分解を促進するアンチザイムという蛋白質がポリアミン輸送も負に調節していることが明らかとなった。すなわち、アンチザイムcDNAをマウスFM3A細胞にtransfectすると、ODC量の減少と共に、ポリアミン輸送活性も減少した。
