受容体非介在性の生体内脂質運搬能を有するシクロデキストリン誘導体の構築

研究概要

本年度は、α-、β-、およびγ-シクロデキストリンの非晶質誘導体の硫酸エステル化体あるいはスルホアルキル化体を用いた受容体非介在性の脂質運搬システムの構築に関する検討を行い、以下の知見を得た。
1)超遠心法により調製した血清脂質分画(カイロミクロン、超低比重リポ蛋白(VLDL)、低比重リポ蛋白(LDL)、および高比重リポ蛋白(HDL))と上記シクロデキストリンとの相互作用を水系ゲル瀘過カラムを装着した高速液体クロマトグラフィーを用いて検討した結果、メチル化シクロデキストリンはHDLと、硫酸化シクロデキストリンはカイロミクロンやVLDLと強く相互作用することが明らかとなった。
2)硫酸化シクロデキストリンをラットに静脈内投与すると、二相性の血中消失パターンを示し、その大部分が尿中に排泄された。その際、硫酸化シクロデキストリンの定常状態における分布容積や全身クリアランスは親水性シクロデキストリンやイヌリンよりも小さいことから、硫酸基の導入によってシクロデキストリンの体内分布や腎排泄が制限されたものと推察された。
3)硫酸化シクロデキストリンをラットやウサギに非経口的に大量投与した場合の臓器障害性や局所組織障害性は、天然シクロデキストリンや他の硫酸化多糖類よりも低かった。
4)高コレステロール食を摂取した外因性動脈硬化症モデルおよび肝臓のLDL受容体が遺伝的に欠損した先天性動脈硬化動物モデルを用いた検討において、硫酸化シクロデキストリンを静脈内に長期間投与すると、血中脂質レベルに顕著な変化はみられなかったが、動脈硬化病巣の退縮が観察された。
本研究で得られた知見は、新しいタイプの抗動脈硬化薬の研究開発や血管形成術後の再狭窄防止策を確立する際の有益な基礎資料となるものと考えられる。
