研究概要	マウスT/t遺伝子複合体は第17番染色体に位置する遺伝領域で、胚発生、神経機能、生殖細胞機能等に関連する突然変異が多数マップされており、またマウスの主要組織適合性遺伝子複合体(MHC)であるH-2領域全域を含むことが知られている.我々は、マウスMHCのK領域のごく近傍に初期着床胚にて致死をおこすt^<w5>発生致死変異が位置することを遺伝学的に見出だし,コスミド、およびYACクローンにより約800kbのゲノム領域をクローニングした.マウスの交配実験によりt^<w5>変異はH-2K遺伝子に強く連鎖しており、最大でも1cM以内にあることがわかっていたが、マイクロサテライトプローブによる詳細なマッピングの結果、突然変異を含むゲノム領域がほぼカバーされていることが判明した.t^<w5>変異責任遺伝子単離を最終目標に、この領域の未知遺伝子を我々の開発した高効率液相スクリーニング法により探索し、これまで22の新規遺伝子を発見した.遺伝子機能を指標とした原因遺伝子クローニングの新しい手法として、YAC DNAを導入したトランスジェニックマウスの作製も試みており、H-2K領域約650kbをカバーするYAC DNAを損傷なく単離、精製しマイクロインジェクション法によりマウス受精卵に導入し3系統のトランスジェニックマウスを得ている.これらのマウスにおいて導入DNAがどのようにゲノムに組み込まれているかを解析中である.
