痴呆モデルとしての時刻認知障害動物モデルの作製と改善薬の開発に関する研究

研究概要	本年度は、夜間徘徊の動物モデルつまり時刻認知障害の動物モデル作製に努めた。その結果【.encircled1.】外界の明暗情報の障害動物モデルの作製。老齢動物を恒常明の条件で飼育したところ、運動量の低下とともに、視交叉上核の神経活動のサーカディアンリズムの低下がみられた。つまり老齢動物の運動量、飲水量などのサーカディアンリズムの周期は短くなり、位相変化は弱く、振幅は小さくなった。さらにこのことに呼応して体内時計の視交叉上核神経のニューロン活動のサーカディアンリズムにも異常があることがわかった。【.encircled2.】社会的同調障害による時刻認知障害の動物モデル。老齢動物に毎日一定時刻に給餌やメタンフェタミン投与行なっても、時刻認知の形成に伴う運動量の増大が若いラットではみられるが、老齢動物では障害されていることがわかった。【.encircled3.】これらの時刻認知障害の動物モデルが迷路学習の空間認知障害モデルとどのように異なっているかにつき調べ、時刻認知障害の動物モデルの特性を明らかにしようと試みた。つまり、種々の薬物の薬理作用が異なるか否について調べた。NMDA受容体拮抗薬で給餌制限およびメタンフェタミン投与ラットのいずれの時刻認知も著明に障害したが、抗コリン薬ではほとんど障害が見られず、この点は迷路学習の場合とは異なっていた。
