PCRを用いた国内産 Verticillium属菌の分類

研究概要

PCRを利用したRAPD(Random Auplified Polymevphic DNA)法はRFLP分析法に比べ、簡易かつ迅速にDNAの変異を検出できる。そこでまず、Verticillium clahliaeの5つの菌群(A:ナス系、B:トマト系、C:ピ-マン系、D:アブラン科系、E:病原性不明株)の代表となる菌株(農研センター萩原氏より分離)のRAPDの解析を試みた。それぞれの菌株のDNAを抽出し、その各10ngを鋳型として、dNTP200muM、プラズマ-0.2muM、Taqポリメラーゼ2.0unitsを加えた反応液を94℃1分、36℃1分、72℃2分の条件で、40サイクルのPCR判翁を行なった。プライマーには任意の配列からなる10merの20種類の合成プライマー(オペロン社.kit A)を用いた。その結果、20種類のプライマーのうち12種類で増幅されたDNA断片に多型が認められ、供試した5つの菌株の相互の類別が可能であった。次に、DNAの多型を示すバンドの多かったプライマー(Opa-04、10、12)を3種類用いて、V.dahlin(16菌株)、V.alboratrn(4菌株)のRAPDの解析を行ない、バンドの有無でクラスター分析をした。その結果、V.dahlineとValbo atrnはそれぞれクラスターに別れた。V.dahline内も、若干の例外は認められたが、同一菌群でサブクラスターを形成する傾向にあり、本法がVerticillium属菌の分類に有効であることが示された。しかし、プライマー数と菌株数を増やすことにより、さらなる解析が求められる。
