神経伝達物質による軸索輸送の制御機構と細胞内セカンドメッセンジャーの関係

研究概要

近年軸索輸送に関する研究は、コンピュータ方式によるビデオ増感微分干渉顕微鏡の開発により急速に進歩を遂げている。ことに軸索輸送の生化学に関する研究の発展は著しい。しかし培養神経細胞を用いて、生理的条件下において、軸索輸送の制御機構についての詳細な検討は、一般にされていない。上記顕微鏡を用いて約1万倍の倍率で、軸索輸送を直接観察しながら実験を行ってきた結果、前々年度単離培養した交感神経細胞にアセチルコリン(ACh)を投与し、軸索輸送の、速度、数量とも減少する、可逆的抑制が起り、この現象にはムスカリンリセプターが関与しGi蛋白をセカンドメッセンジャーにして起ることをつきとめ報告した(Bain Research,1992年)(Biomedical Research, 1991年)。その後軸索輸送の促進因子に関する研究を行い、同細胞を用いた場合アドレナリンの投与により軸索輸送の速度、数量共に増加が認められ、詳細に検討した結果、この現象はbeta2リセプターを介し、アデニル酸シクラーゼを活性化しcAMPの増加により引き起こされることを発見報告した(Bain Research,1994 in press)。以上の結果より軸索輸送の制御機構には、神経伝達物質が関与し、各々のリセプターお介し、夫々のG蛋白を活性化した後cAMPの増域により行われることがほぼ明らかとなった。更に現在、神経細胞内のリセプターの局在と軸索輸送の制御機構の関連を研究するためにマイクロマニピュレータとインジェクターを上記顕微鏡に接続し局所に薬物投与を行った時の軸索輸送の変化即ち、神経細胞の細胞体、神経線維、成長錐の各部位ごとにACh,アドレナリンを投与し観察を行っている。また、並行して免疫組織化学的手法を用いリセプターの同定と神経細胞内における局在についても現在検討中である。来年度は、更に進んで軸索輸送の制御機構について研究を推進する予定である。
