rho p21 GTP結合蛋白質による細胞骨格の制御機構とその病態

研究概要

本研究では、Rho低分子量G蛋白質の機能と活性化機構について検討した。増殖因子を細胞に作用させると、アクトミオシン系を介して細胞膜のラッフリングや細胞運動が誘起される。RhoからのGDPの解離を抑制することによりRhoの活性化を阻害するRho GDIをKB細胞へマイクロインジェクションしたところ、インスリンやHGFにより誘起される細胞膜のラッフリングが阻害された。インスリンにより誘起されるラッフリングの阻害はGTPgammaS-Rac(RacはRhoに類似した低分子量G蛋白質)により、HGFにより誘起されるラッフリングの阻害はGTPgammaS-Rhoによりそれぞれ回復した。また、Rho GDIをケラチノサイト308R細胞へマイクロインジェクションしたところ、HGFにより誘起される細胞運動が阻害された。この阻害はGTPgammaS-Rhoにより回復した。-方、私共はRhoに作用する活性制御蛋白質としてGDP解離促進蛋白質(Smg GDS)とGDP解離抑制蛋白質(Rho GDI)を見出してその性状を解析している。最近、Dblがん遺伝子産物がRhoに類似したCdc42のGDSとして作用することが報告されている。本年度は、DblがRhoのGDSとして作用するか否かを検討した。大腸菌で発現して精製したDblはRhoファミリーに属するRho、Rac,Cdc42のGDP/GTP交換反応を促進した。私共のこれまでの解析から、Smg GDSは翻訳後修飾を受けているRhoにのみ作用することが明らかとなっていたが、Dblも翻訳後修飾を受けているRhoに対してより高い活性を示した。さらに、DblはSmg GDSの場合と同様にRho GDIと複合体を形成しているRhoには作用できず、したがって、RhoからのGDPの解離を促進できなかった。以上、Rhoの機能と活性化機構の詳細がかなり明らかになり、当初の研究目的はほぼ達成することができた。
