c-fos遺伝子発現を指標とした歯の移動に伴う中枢投射領域の変化の解析

研究概要	近年歯科臨床において、歯科矯正は重要となってきたが、歯の移動に伴う痛みは深刻な問題の一つである。本研究では、外部からの侵害刺激の際に中枢神経系に発現するとされる c-fos遺伝子の発現を指標として、歯の移動に伴う中枢神経系である二次ニューロンに認められる変化を、免疫組織科学的手法により解明することができた。実験には、ラットの上顎片側臼歯歯間にゴム片を挿入し、歯の移動モデルとした。ゴム片挿入から 0、1、2、4、8、12、24時間後に動物を潅流固定し、脳幹を摘出、凍結切片を作製した。抗c-fos血清、さらにHRP標識二次抗体を用い免疫染色し、光学顕微鏡にて観察した。c-fos陽性細胞の分布を明らかにし、さらに時間経過にともなうその分布領域の変化と、その細胞数の増減についても明白にした。結果は、以下の通りである。 (1) ゴム片挿入の1時間後から、同側の三叉神経脊髄路核尾側亜核(STNC)の、神経細胞体内の核に限局してc-fos陽性反応を認め、24時間後にはc-fos陽性細胞はすべて消失した。 (2) c-fos陽性細胞の分布は、STNC表層のI層と、II層の外層のそれぞれ正中寄り3分の1に限局していた。 (3) ゴム片挿入と同側のc-fos陽性神経細胞の数は、4時間目に極大に達し、24時間後には消失していた。 (4) 一方、ゴム片挿入の反対側では、 1,2,4時間後にのみ、実験側と同じ部位のSTNCに、c-fos陽性細胞を認めたものの、その数は少なく、分布はまばらであった。以上の結果より、c-fosは歯の移動に伴う痛みに関係があることが示された。さらに、STNCは、痛みの調節に関連が深いことも明白となった。
