DNAと薬物の相互作用に関するx線構造化学、および構造活性相関研究

研究概要	昨年から本年にかけてスペルミジンとZ-DNA複合体結晶のx線結晶構造解析の結果を詳細に検討を行い、論文にまとめたこの論文は現在、投稿前段階である.またN-(2-aminoethyl)-1,4-diaminobutaneとZ-DNA複合体結晶、スペルミジンとZ-DNA複合体結晶および、スペルミンとZ-DNA複合体結晶の構造の構造比較、ポリアミンのDNAに対する役割、およびポリアミンの鎖長の違いによる相互作用様式の検討を行い、現在投稿準備中である.その他、DNAと相互作用し抗菌活性、植物成長疎外活性を示すのではないかと考えられていたジアチン化合物、ヒノキチオール化合物、ユ-デストミン化合物などのx線結晶構造解析、およびDNAとの相互作用活性、構造活性相関などを調べた結果、ヒノキチオール化合物、ジチアン化合物はDNAと相互作用して活性を示すのではないことがわかった.これらの現象に関してはヒノキチオール化合物、ユ-デストミン化合物に関してはすでに論文審査も終了し現在印刷中である.また、ジチアン化合物に関しては構造の面からと構造活性相関のめんから論文を公表し現在審査中である.またDNAと相互作用し制癌活性を示すシスプラチン化合物4種類のx線結晶構造解析に成功し制癌活性発現機構および、詳細な構造を検討し投稿準備中である.これらのシスプラチン化合物とDNAの複合体の結晶化も現在行っている.この構造解析が成功すれば制癌活性発現機構など、より詳細な事実が明らかになることと考えられる.またコンピュータを用いたDNAの解析では、われわれがすでにx線結晶構造解析によって明らかにしている左巻きd(CG)3とコンピュータで構築した右巻きd(CG)3とを熱振動および力場計算を用いたダイナミックシュミレーションを試み、なぜ塩濃度を上げるとB-Z転移が起こるのか、熱をかけるとおなじ転移が起こるのか、またポリアミン濃度によってもおなじ転移が起こるのか、Z-DNAが生体内でどのような役割を果たしているのかを検討している.インターカレーターをいれたDNAがダイナミックシュミレーションを行うことによってどのような位置に安定に存在しえるのかも今後検討していきたいと考えている.
